Identification of genes associated with perianth senescence in Daffodil (Narcissus pseudonarcissus

چکیده
We used a PCR-based subtractive procedure to isolate genes that increased in abundance in the perianth of incipiently senescent daffodil (Narcissus pseudonarcissus L. ‘Dutch Master’) flowers. Using RNA gel blot analysis we confirmed that 18 of the transcripts were senescence-associated. A number of these have homologs in previously-studied senescing floral and leaf tissue systems (cysteine proteases, serine proteases, endonuclease, vacuole processing enzyme). Some showed homology to genes encoding proteins with transport functions (auxin efflux carrier protein, low affinity nitrate transporter), others to enzymes that may have a role in cellular signaling (RSH-like protein). A surprising number of the isolated genes either showed no homology with previously-reported sequences or were sequences with no known function. A strong association of some of the transcripts with senescence was also indicated by their enhanced accumulation in the perianth of detached daffodil flowers, which senesce earlier than attached flowers. # 2002 Elsevier Science Ireland Ltd. Senescence-associated Gene Expression in Narcissus ‘dutch Master’
Flower senescence is a highly controlled developmental event that culminates in the death of the floral organs. Most research on flower senescence has focused on the perianth, since it typically determines the commercial life of the flower. The mechanism by which the perianth dies differs between species: in some flowers, wilting is the primary symptom of senescence whereas in others, the peria...
متن کاملComparison of the Polypeptide Complement of Different Plastid Types and Mitochondria of Narcissus pseudonarcissus
The protein complement of chloroplasts, prochromoplasts, chromoplasts, and mitochondria of the daffodil (Narcissus pseudonarcissus) has been investigated comparatively by gel electro phoresis. Mitochondria do not share common proteins with plastids. On the other hand, there is a broad overlap of the protein complement of the three plastid types investigated, in spite of their different fine st...
متن کاملCloning and Characterization of a Norbelladine 4′-O-Methyltransferase Involved in the Biosynthesis of the Alzheimer’s Drug Galanthamine in Narcissus sp. aff. pseudonarcissus
Galanthamine is an Amaryllidaceae alkaloid used to treat the symptoms of Alzheimer's disease. This compound is primarily isolated from daffodil (Narcissus spp.), snowdrop (Galanthus spp.), and summer snowflake (Leucojum aestivum). Despite its importance as a medicine, no genes involved in the biosynthetic pathway of galanthamine have been identified. This absence of genetic information on biosy...
متن کاملFunctional analysis of a RING domain ankyrin repeat protein that is highly expressed during flower senescence.
A gene encoding a RING zinc finger ankyrin repeat protein (MjXB3), a putative E3 ubiquitin ligase, is highly expressed in petals of senescing four o'clock (Mirabilis jalapa) flowers, increasing >40,000-fold during the onset of visible senescence. The gene has homologues in many other species, and the Petunia homologue is strongly up-regulated in senescing Petunia corollas. Silencing the express...
متن کاملA Simple and Rapid Method for Isolating High Quality Rna from Flower Petals
A simple RNA isolation method based on hot borate and proteinase K was used to extract high-quality RNA from petals of daffodils (Narcissus pseudonarcissus L. 'Dutch Master'). The method was adapted from the isolation technique described by Wan and Wilkins (1994). A single isopropanol precipitation and ethanol wash replaced all the second day steps of the Wan and Wilkins procedure. The RNA isol...
